The Kloros Guild Tower will be locked from the inside. You will have to defeat the enemy to open it. This enemy is inside the tower, so you will have to find another way in. To do this, grapple up to the rock pillar on the left (you unlock the grapple after the first mayor boss), then jump to the closer pillar to finally jump through the balcony. Once inside go to the bottom floor to defeat the enemy.
